Exploring the effect of secretome of platelet-mesenchymal stem cells spheroids on angiogenesis and healing in chronic wounds : DST-SERB ongoing

in- vivo evaluation of 3D bioprinted skin tissue construct

3D Bioprinting of Skin tissue construct: DST-TRC, PI,Hydrogel as bioink was synthesized, characterized and optimized for printability. Dermal epidermal culture was optimized, constructs are cultured and analysis of functional markers are ongoing.

Development of the assay platform and sensing device for Prothrombin Time/INR monitoring DST-TRC funded, PI, Technology is transferred to Agappe Diagnostic Pvt Ltd, Device improvement is ongoing for increasing the sensitivity of the device.

3D Bioprinting of Liver, DST-TRC Co-I, 2016-19. Cellular studies for the collagen based bioink formulated as well as explored the differentiation of ADMSCs to hepatic like cells in order to establish an alternative cell source for the hepatic regeneration. Cells functionality and viability was proven based on the live dead assay and functional assay like cyto P-450, albumin secreation, cardiogreen uptake, glycogen storage and urea production.

Development of Leukodepletion filter, DST-TRC, Co-PI, 2017-20. Different batches of filters were tested for blood filtration and best one with 100% depletion was identified and is taken forward for the further studies. Filter material was modified to improve filtration efficiency.

Monocyte proteome profiling in diabetic subjects. Proteomic profiling of monocytes from diabetic subjects were done and analyzed for the proteins causing cardiovascular diseases in diabetic subjects.

Coronary Artery Disease in the Young: KSCSTE funded, Co-I 2010-14: Platelet activation status in young patient was evaluated and platelet monocytes interaction studies were done using CD62 and CD14. Inflammatory market analysis in young cardiac patient was also carried out. Monocytes phenotypes were also analyzed using CD14 and CD16 antibodies using flowcytometry. Two publications as co author and three conference paper as co author was the outcome of this project.

Quantum dots as a drug carrier in cardiovascular diseases . DST funded, Co-PI ,2011-2014 : CdSe/ZnS based quantom dots were synthesized, characterized and tagged with specific antibody. Encapsulation of the targeted system and effect on endothelial cells were studied in-vitro. Biocompatibility studies on the synthesized QDs were also carried out. QDs were found to be specific. Three papers were published and one paper was presented in an International conference and was published in the proceedings.

Role of platelet proteins on the Endothelial Cells and Smooth Muscle Cells proliferation: SCTIMST funded,PI, 2012-14. Platelet proteome was isolated and effect of platelet proteome on endothelial cells and smooth muscle cell proliferation was evaluated in-vitro. Activated platelet proteome was assayed to analyze the variation from the resting healthy platelets.

Do platelet in patients with type II diabetes secretes proteins that can activate aortic endothelial cells funded by KSCSTE , PI, 2014-17. Proteomic profiling of twenty subjects from each group (diabetic and healthy) were done. Data was analyzed by using PD quest 2D gel analysis software. Significant differences between the groups were observed. Mass spectrum analysis was done for the samples. Western blot analysis was done for PF4, beta thromboglobulin, secretogranin, cyclophilin A. Mass pack analysis of data revealed the up-regulation of inflammatory and oxidative stress pathways in the diabetic subjects. One paper was published as a outcome of this project. Two man power was trained and two conference paper was presented and two workshop was attended.

3D Bioprinting of skin tissue construct: Dual layer skin tissue construct were printed and analyzed in-vitro and in-vivo. In-vitro studies showed functional skin markers and the preliminary in-vivo studies demonstrated better wound healing.

Mesenchymal stem cells and activated platelets, role of secretome in wound healing : MSCs will be induced by activated platelets. MSCs secrtome will be analyzed and wound healing efficiency will be analyzed.

Development of the PT INR assay platform : A novel device is developed to measure prothrombin time . Initial experiments showed exciting results. Research is in progress to achieve an accurate point of care device
